Service Description: we provide services to use pronuclear DNA microinjection method to generate transgenic rabbits. Normally this method is ideal for the insertion of new genes to the rabbit genome, for example, to generate humanized transgenic rabbits, or to use rabbit as a bioreactor to produce certain therapeutic proteins.

The whole package service includes animal purchase, animal care, hormone treatment, superovulation, oocyte harvest, DNA microinjection, embryo culture, embryo transfer, near natal care and sampling of kits. We expect our customers to provide high quality DNA construct, to perform genotype confirmation of the animals, and to transport animals after completion of the project. 

 

Normally customers provide DNA construct for microinjection. We recommend customers test their construct in a smaller animal (mostly in mice) before being used in the rabbits.

 

1. MI-ET (DNA microinjection & embryo transfer)
This is the main service package; the package includes all service items described below, and is charged as one MI-ET service. It includes:


(1) The work to collect zygote stage rabbit embryos, perform DNA microinjection, in vitro culture the embryos, prepare the recipient animals, and perform embryo transfer
(2) Animal pregnancy and birth care
(3) C-section on recipients when necessary

(4) Baby rabbit care till the age of 35 days old

 

Note: With approximately 300 embryos MIed, it is expected that 2-3 positive animals will be generated. However, transgenic rate vary depending on the transgene construct used.

 

2. EAR-SMP (ear sampling)
This service includes collecting a 5mm diameter skin tissue from one ear of a young rabbit normally at the age D35 to D45. This is normally for genotyping assays.


The customer is responsible for any shipping charges.

 

3. BLOOD-SMP (blood sampling)
This service includes collecting blood from ear vein from a rabbit normally older than 6 weeks.


The customer is responsible for any shipping charges.

 

4. PCR-GENO (PCR genotyping)
This service includes performing PCR assay to determine the genotype of the animals.

 

The customers will provide primer sequence.

 

5. PER-DIEM
We charge per diem for any rabbit in the project that is older than 35 days old.

 

6. BRED (breeding of a pair of rabbits)
We charge the breeding program.

 

7. RAB (rabbit purchase)
We charge extra male and female rabbits other than donors and recipients.

Advantages of Rabbit Model

Although the study of many human diseases has benefited significantly from the use of mouse models, small rodents do not always accurately reflect human physiology


Take the cardiovascular system as an example. The length of the mouse’s cardiac cycle is only one-tenth of that of the human. Indeed, murine and human hearts differ in several fundamental facets, and these differences are reflected at the molecular levels.  For example, the cardiac sarcomere in the mouse consists of the "fast" myosin heavy chain (MHC) isoform a(a-MHC), whereas the "slow" b-MHC is the dominant isoform in the human ventricles.  In addition, the high-density lipoproteins (HDL) are the predominant form of lipoproteins in the mouse plasma in contrast to the low-density lipoproteins (LDL) that are predominant in humans, conferring resistance to diet-induced atherosclerosis. Furthermore, mice are too small for any practical applications such as repeated surgeries, monitoring and/or measurements, which are essential for cardiovascular research and practice.

 

The rabbit would be an ideal choice (Table 1), considering the following: 1) Rabbit ventricles are similar to humans’ and both a- and ß-MHC isoforms are expressed with ß-MHC being the predominant isoform in adults; 2) Rabbit lipoprotein metabolism is very similar to that of humans (e.g., LDL-mammals, same as humans), but very different from mice (HDL-mammals); 3) Cholesteryl ester transfer protein (CETP), which plays a central role in the atherosclerotic process, is abundant in both human and rabbit plasma but absent in the mouse; 4) Like humans, rabbits are very susceptible to diet-induced atherosclerosis.

 

Comparing to mice, rabbits are phylogenetically closer to human. Because of the anatomical, physiological, genetic and biochemical similarities between the rabbit and the human, this species is preferentially used in pulmonary, cardiovascular and metabolic studies, including airway obstructive disease, embolic stroke, arteriosclerosis, cholera, cystic fibrosis, neoplasia, diabetes, acute respiratory distress syndrome, malignant lymphoma, acquired immunodeficiency syndrome and hypercalcemia of malignancy.  

 

As a classical experimental animal species, rabbit has several other advantages over some other animal species (e.g., monkey, pigs). It has a short gestation period (30-31 days), large litter size (4-12/litter) and can be housed conveniently in an indoor facility.   In addition to the use as an animal model, rabbit is also used for drug screening, antibody production, and the production of therapeutic proteins (bioreactors). Some proteins can be properly synthesized in rabbit milk, but not in other animal systems (e.g. cattle or goats) due to the post translation processing.
